Any container will do the trick. The main reasons for putting them in a 
plastic cups is:

1. you don't have to worry about getting the glasses back for washing.
2. The plastic cups are cheap enough to make several dozen shots in advance. 
3. Some plastic cups come with covers. This makes it easier to stack, store,   
  and label.
4. Easier to eat if you can squeeze the shot out of the cup.

It would be funny watching drunk people trying to suck jello shots out of a 
plastic test tube.  lol
