In a quart bottle (32 oz) I would figure at an ounce per shot. 32 shots in a 
bottle.  In a fifth there are (at one ounce per shot) 25.6 shots.

You factor in everything: liquor, ice, garnishes, and mixes such as juice and 
sodas.  I would not factor in labor.  I would take the total cost of the above 
ingredients and multiply by 3.  2/3 covers your cost (ingredients, labor, and 
other costs) and the 1/3 is pure profit.

I was told when I took a business course that this is how it is figured if you 
own a store in which you are selling a product, however it may be different 
with the liquour/bar industry.

Let me know what the cost would be to make this drink:  

Sea Breeze

1 oz Vodka
1 oz cranberry juice
1 oz Grapefruit juice
3 oz of ice.
Garnish: lime wheel

In a 6 oz highball glass add all ingredients.  Garnish: lime wheel

I went to a bar over the weekend and had a:

Pearl harbor: 1 oz midori.  1/2 oz vodka and 1.5 oz pineapple juice (3 oz 
ice).  it was $3.50 + TIP.  No garnish.
